Retrograde Transport

Retrograde transport is the directed movement of materials from a cell’s periphery toward its central regions, supporting organization, communication, and recycling within eukaryotic cells. Molecular motors such as dynein carry vesicles, endosomes, and damaged organelles along microtubules toward the microtubule-organizing center, often using adaptor proteins to select cargo. In neurons, this pathway moves signaling endosomes from axon terminals to the cell body, where signals can influence gene expression and survival. Retrograde transport also returns proteins from the Golgi apparatus to the endoplasmic reticulum and can be exploited by toxins and viruses, making it important in cell biology, disease research, and therapeutic development.

Source: Keefe, K. M., et al. Direct Injection of a Lentiviral Vector Highlights Multiple Motor Pathways in the Rat Spinal Cord. J. Vis. Exp. (2019).This video demonstrates the injection of a highly efficient retrograde transport (HiRet) viral vector into the rat spinal cord to trace neuronal pathways. The virus has glycoproteins for synaptic entry and a fluorescent protein coding gene. The needle is inserted into the spinal cord, and the virus suspension is administered at a controlled rate.

Retrograde Fluorescent Labeling Allows for Targeted Extracellular Single-unit Recording from Identified Neurons In vivo

2013

Retrograde transport of fluorescent dye labels a sub-population of neurons based on anatomical projection. Labeled axons can be visually targeted in vivo, permitting extracellular recording from identified axons. This technique facilitates recording when neurons cannot be labeled through genetic manipulation or are difficult to isolate using 'blind' in vivo approaches.
