Single Cell Assay

Single-cell assays are experimental methods that measure molecular, genetic, or functional properties in individual cells rather than in pooled populations, revealing biological variation that bulk analyses can obscure. These assays typically isolate cells or assign measurements to single-cell compartments, then detect features such as proteins, transcripts, metabolites, or cellular responses through imaging, fluorescence-based detection, or sequencing. In biology, single-cell assays help identify rare cell populations, define cell states, trace developmental changes, and characterize disease-associated heterogeneity. Their ability to link molecular profiles with individual-cell behavior supports more precise studies of tissues, immune responses, cancer, and therapeutic effects.

Quantitative High-throughput Single-cell Cytotoxicity Assay For T Cells

We describe a single-cell high-throughput assay to measure cytotoxicity of T cells when incubated with tumor target cells. This method employs a dense, elastomeric array of sub-nanoliter wells (~100,000 wells/array) to spatially confine the T cells and target cells at defined ratios and is coupled to fluorescence microscopy to monitor effector-target conjugation and subsequent apoptosis.

In vitro spheres assays are commonly used to identify cancer stem cells. Here we compare single with multi cell-based spheres assays. The more laborious single cell-based assays or methylcellulose supplementation give more accurate results while multi cell-based assays performed in liquid medium can be highly influenced by cell density.
