Whole-mount Quantification

Whole-mount quantification is the measurement of cellular, structural, or molecular features across an intact biological specimen rather than in thin tissue sections. Researchers typically label the feature of interest, image the entire specimen, and analyze the resulting data to determine values such as object number, size, distribution, or signal intensity while preserving spatial relationships. This approach supports studies of development, tissue organization, morphology, and disease-related changes in model organisms and organs. By combining specimen-wide visualization with reproducible image analysis, whole-mount quantification enables comparisons between experimental conditions and helps link local biological events to patterns across complex tissues.

Three-dimensional Quantification of Intestinal Mucus Using Whole-mount Tissue Imaging

2025

This study presents a 3D imaging method using whole-mount intestinal tissues and multi-photon microscopy to quantify secreted mucus, enabling precise volumetric analysis and visualization of mucus dynamics in response to stimuli like carbamoylcholine chloride.

Whole Mount Immunofluorescence and Follicle Quantification of Cultured Mouse Ovaries

Here, we present a protocol to quantify follicles in cultured ovaries of young mice without serial sectioning. Using whole organ immunofluorescence and tissue clearing, physical sectioning is replaced with optical sectioning. This method of sample preparation and visualization maintains organ integrity and facilitates automated quantification of specific cells.

Whole-Mount Dissection of the Vestibular Complex and Quantification of Vestibular Ganglion Neurons in Mice

2026

We present a new dissection method to extract the vestibular ganglion and all five vestibular organs (utricle, saccule, and three cristae: lateral/horizontal, anterior, and posterior) as one whole-mount vestibular complex. Confocal imaging allows visualization of vestibular neuron cell bodies for quantification, as well as neuronal projections to the sensory...

Whole-Mount In Situ Hybridization

2023

Whole-mount in situ hybridization (WMISH) is a common technique used for visualizing the location of expressed RNAs in embryos. In this process, synthetically produced RNA probes are first complementarily bound, or "hybridized," to the transcripts of target genes. Immunohistochemistry or fluorescence is then used to detect these RNA hybrids, revealing spatial and temporal patterns of gene expression. Unlike traditional in situ hybridization techniques, which require thin tissue sections whose...
