C26 Carcinoma

C26 carcinoma is a transplantable murine colon carcinoma model used to study tumor growth, tumor-host interactions, and cancer-associated systemic effects. Researchers typically implant C26 cells into syngeneic BALB/c mice, allowing the tumor to develop in an immunocompetent setting while preserving relevant immune responses. As the tumor grows, inflammatory signaling, including interleukin-6 production, can contribute to cachexia characterized by weight loss, muscle wasting, and metabolic changes. This model supports investigation of cancer biology, mechanisms of tumor-induced tissue deterioration, and potential treatments targeting tumor progression, inflammation, or cachexia, making it valuable in preclinical cancer research.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Coculture System with an Organotypic Brain Slice and 3D Spheroid of Carcinoma Cells

The organotypic brain slice coculture with carcinoma cells enables visualizing morphological changes by fluorescence as well as bright field (video) microscopy during the process of carcinoma cell invasion of brain tissue. This model system also allows for cell exchange and replenishment approaches and offers a wide variety of manipulations and analyses.

Generation of an orthotopic mouse model of anaplastic thyroid carcinoma is described here. This technique employs surgical placement of human anaplastic thyroid cancer cells into the thyroid of immunodeficient mice, thus creating a more clinically relevant setting to study disease progression as well as screen innovative therapeutic interventions.
