Cancer Cell Isolation

Cancer cell isolation is the process of separating malignant cells from tumors, blood, or other biological samples so they can be studied as distinct populations. It typically combines mechanical or enzymatic tissue dissociation with filtration, density-based separation, or marker-guided enrichment, while controlled handling helps preserve cell viability and molecular characteristics. In cancer research, isolated cells support analyses of morphology, gene expression, drug sensitivity, tumor heterogeneity, and interactions with the surrounding microenvironment. Reliable isolation also enables primary cultures and downstream genomic or functional assays, improving the connection between patient-derived material and experimental models.

Cancer stem cells (CSCs) have been identified in a number of malignancies. In this protocol we describe a flow cytometric method utilizing aldehyde dehydrogenase activity and CD44 and CD24 expression to isolate CSCs from human pancreatic adenocarcinoma xenografts. These viable cells can then be used in functional and analytical studies.
