Fibroblast Collagen Matrix

A fibroblast collagen matrix is a three-dimensional extracellular environment produced by fibroblasts, the connective-tissue cells that synthesize and organize collagen, and it provides a useful model of tissue structure in cancer research. Fibroblasts secrete collagen and other matrix components, assemble collagen fibers, and remodel the network through contraction, enzymatic turnover, and crosslinking, which alters matrix organization and stiffness. These changes can influence cancer-cell adhesion, migration, invasion, signaling, and response to therapy. In laboratory models, fibroblast-derived matrices help researchers investigate tumor–stroma interactions and evaluate how the extracellular matrix contributes to tumor progression and treatment resistance.

In order to understand the mechanisms that can alter cell migration, we present a planar gradient diffusion system to investigate chemotaxis in a 3D collagen matrix, which allows one to overcome modern diffusion chamber limitations of existing assays.

Explants from the midbrain dopamine system and striatum are used in a collagen matrix assay for the in vitro analysis of mesostriatal and striatonigral pathway development. In this assay axonal outgrowth and guidance can be manipulated and quantified. It can also be modified for assessing other regions or molecular cues.
