Phosphopeptide Isolation

Phosphopeptide isolation is a phosphoproteomics technique that enriches peptides carrying phosphorylated amino acids from complex protein digests, enabling researchers to examine cell signaling and regulatory changes. The method typically uses metal affinity or metal oxide chromatography, in which phosphate groups bind selectively to immobilized metal ions or titanium dioxide under controlled chemical conditions, while unmodified peptides are removed during washing. In cancer research, isolated phosphopeptides support mass spectrometric mapping of kinase activity, signaling pathways, and tumor-associated phosphorylation changes. These data can reveal mechanisms of malignant growth, identify candidate biomarkers, and help evaluate potential therapeutic targets.

Proteomic analysis of any cell type is highly dependent on both purity and pre-fractionation of the starting material in order to de-complexify the sample prior to liquid chromatography mass spectrometry (MS). By using back-flushing techniques, pure spermatozoa can be obtained from rodents. Following digestion, phosphopeptides can be enriched using TiO2.

Post-translational modifications (PTMs) change protein structures and functions. Methods for the simultaneous enrichment of multiple PTM types can maximize coverage in analyses. We present a protocol using dual-functional Ti(IV)-immobilized metal affinity chromatography followed by mass spectrometry for the simultaneous enrichment and analysis of protein N-glycosylation and phosphorylation in pancreatic tissues.
