Spheroid Formation Assay

The Spheroid Formation Assay is a three-dimensional cell-based method that measures the ability of cells to aggregate and grow as compact spheroids, providing a model more representative of tumor architecture than conventional two-dimensional cultures. Under nonadherent or low-attachment conditions, cancer cells cluster through cell-cell adhesion and extracellular matrix interactions, while limited substrate attachment promotes formation of dense, rounded structures. Researchers use spheroids to assess tumor cell self-renewal, proliferation, invasion, drug response, and resistance under physiologically relevant conditions. The assay supports cancer biology, therapeutic screening, and evaluation of tumor heterogeneity, helping bridge findings from cell culture models toward in vivo research.

Spheroid Assay to Measure TGF-β-induced Invasion

An assay to quantitatively measure Transforming Growth Factor (TGF)-β-induced invasion in 3-dimensional collagen gels is described. This assay takes advantage of the MCF10A series of cell lines, which represent different stages of breast cancer development. This method can be adopted to be used with other cell lines and might be used to investigate other potential activators or inhibitors of invasion.

The uneven loss of medium from microtiter plates affects the reproducibility of uniform multicellular tumor spheroid formation. Improving culture conditions to reduce significant medium loss will improve the reproducibility of spheroid formation and the results of spheroid-based assays using the liquid-overlay technique.

An In Vitro Assay to Assess CAR T Cell Cytotoxicity against Tumor Spheroids

This video demonstrates an in vitro assay evaluating the cytotoxic effects of CAR T cells on 3D tumor spheroids expressing fluorescent proteins. This assessment utilizes the Annexin V red fluorescent probe, which binds to exposed phosphatidylserine residues on the outer leaflet of apoptotic cancer cells after interactions with CAR T cells. The red fluorescence emitted by the probe aids in distinguishing these cells from healthy ones emitting green fluorescence.
