Lung Function

Lung function refers to how effectively the respiratory system moves air, exchanges gases, and supports oxygen delivery and carbon dioxide removal, making it central to health and clinical assessment. During ventilation, air flows through the airways into the alveoli, where differences in partial pressure drive oxygen and carbon dioxide exchange across thin respiratory membranes. Clinical tests such as spirometry measure airflow and lung volumes to identify obstructive or restrictive patterns. These measurements help evaluate symptoms, monitor diseases including asthma and chronic obstructive pulmonary disease, assess treatment response, and inform decisions about respiratory care.

Three-dimensional (3D) phase-resolved functional lung (PREFUL) is a functional magnetic resonance imaging (MRI) technique that allows for quantification of regional ventilation of the whole human lung volume, using tidal breathing and contrast-agent free acquisition for 8 min. Here, we present an MR protocol to collect and analyze 3D PREFUL imaging data.

Here, we describe the implementation of phase-resolved functional lung MRI as a contrast-agent-free proton MR technique for the assessment of pulmonary ventilation and perfusion dynamics. Validated and applicable across different field strengths and age groups, it could enhance clinical decision-making in the future by aiding in disease quantification and therapy monitoring.

This paper describes a striaghforward and efficient method of intubating mice for pulmonary function measurements or pulmonary instillation, that allows the mice to recover and be studied at later times. The procedure involves an inexpensive fiberoptic light source that directly illuminates the trachea.
