Embryonic Signals

Embryonic signals are molecular and cellular cues that coordinate how an embryo grows, patterns its tissues, and acquires distinct cell identities, making them central to developmental biology. They act through cell-cell communication, often when signaling molecules bind receptors and activate gene regulatory networks; spatially distributed morphogen gradients can give cells positional information and trigger different developmental programs at defined concentrations. These signals guide processes such as cell differentiation, tissue organization, and organ formation. Understanding their timing and interactions helps explain congenital abnormalities, informs studies of stem cell differentiation and organoids, and supports research in regenerative medicine and developmental disease.

A System for ex vivo Culturing of Embryonic Pancreas

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.

Assaying the Ability of Diffusible Signaling Molecules to Reorient Embryonic Spinal Commissural Axons

This assay assesses the ability of a signaling molecule, here Bone Morphogenetic Protein 7 (BMP7), to reorient commissural axons. An explant of embryonic dorsal spinal cord is cultured adjacent to an aggregate of COS cells secreting the candidate growth factors. Reoriented commissural axons growing within the explant are visualized by immunohistochemistry.

Embryonic stem (ES) cells are undifferentiated pluripotent cells, meaning they can produce any cell type in the body. This gives them tremendous potential in science and medicine since they can generate specific cell types for use in research or to replace body cells lost due to damage or disease. Origin ES cells are present in the inner cell mass of an embryo at the blastocyst stage, which occurs at about 3–5 days after fertilization in humans before the embryo is implanted in the uterus.

Analysis of Coronary Vessels in Cleared Embryonic Hearts

We present a protocol for the analysis of coronary vessels in whole embryonic murine hearts up to E15.5, using standard immunological staining methods followed by optical clearance and confocal microscopy. This technique enables visualization of blood vessels throughout the entire heart without the need for time-consuming analysis of serial sections.
