Induced Trophoblast Stem Cells

Induced trophoblast stem cells are reprogrammed cells that acquire the self-renewing, multipotent properties of trophoblast stem cells, which give rise to the fetal components of the placenta. Their production typically involves exposing differentiated cells to defined transcriptional and culture conditions that reset cell identity and activate trophoblast lineage programs, allowing continued proliferation while retaining the capacity to differentiate into specialized trophoblast subtypes. In developmental biology, these cells provide a controllable model for studying placental development, implantation, and cell fate regulation. They may also support research into pregnancy complications, disease mechanisms, and potential regenerative or screening applications.

In this video, we demonstrate the isolation of mouse blastocysts and the derivation of trophoblast stem cells from blastocysts. We also describe conditions for maintenance of the stem cell property as well as induction of differentiation in culture.

Here, we present a protocol to efficiently generate human trophoblastic cells from human pluripotent stem cells using bone morphogenic protein 4 and inhibitors of the Activin/Nodal pathways. This method is suitable for the efficient differentiation of human pluripotent stem cells and can generate large quantities of cells for genetic manipulation.

Isolation of Primary Mouse Trophoblast Cells and Trophoblast Invasion Assay

In this protocol, we describe the dissection of placentae from the mouse on pregnancy d10.5, followed by isolation of trophoblast cells using a Percoll gradient. We then demonstrate use of the isolated cells in a matrigel invasion assay.
