Optic Cup Removal

Optic cup removal is a microsurgical technique in developmental biology that excises the optic cup from an early embryonic eye to examine its role in ocular development. The optic cup, a two-layered structure formed from the optic vesicle, normally interacts with the surface ectoderm and surrounding tissues through reciprocal inductive signals; removing it disrupts these interactions and reveals their developmental contributions. Researchers use this approach to investigate the formation of the neural retina, retinal pigment epithelium, lens, and other eye tissues. Findings clarify how tissue interactions guide organ patterning, differentiation, and regeneration during eye development.

4-Dimensional Imaging of Zebrafish Optic Cup Morphogenesis

This protocol describes an approach for in toto labeling and multidimensional imaging of zebrafish early eye development. We describe labeling, embedding, and four dimensional (4D) imaging using laser scanning confocal microscopy, and considerations for optimizing acquisition of datasets for dissecting mechanisms of optic cup morphogenesis.

We discussed the synthesis of individual graphitic nanocups using a series of techniques including chemical vapor deposition, acid oxidation and probe-tip sonication. By citrate reduction of HAuCl4, the graphitic nanocups were effectively corked with gold nanoparticles due to the chemically reactive edges of the cups.

Optical-resolution photoacoustic microscopy (OR-PAM) is an emerging technology capable of imaging optical absorption contrasts in vivo with cellular resolution and sensitivity. Here, we provide a visualized instruction on the experimental protocols of OR-PAM, including system configuration, system alignment, typical in vivo experimental procedures, and functional imaging schemes.
