Dna Cleanup Step

A DNA cleanup step is a purification procedure that removes unwanted substances from a DNA sample before downstream genetic analysis, improving the reliability of the results. It typically uses selective DNA binding to a silica membrane or magnetic particles, followed by washing to eliminate primers, nucleotides, proteins, enzymes, and salts, then elution in a clean solution. In genetics, DNA cleanup supports PCR, sequencing, cloning, and fragment analysis by reducing contaminants that can inhibit enzymes or interfere with signal detection. Consistent purification helps produce higher-quality templates and strengthens the accuracy and reproducibility of molecular experiments.

For clinical translation, large eye animal models are required to study surgical techniques applicable in patients. Here we present a rabbit model for subretinal surgery geared towards RPE transplantation, which is versatile and cost-efficient.

Here we describe a method for the isolation of hepatic stellate cells from mouse liver. For stellate cell purification, mouse livers are digested in situ and in vitro by pronase-collagenase treatment prior to density gradient centrifugation. This technique yields highly pure hepatic stellate cells.
