Egg Raft Separation

Egg raft separation is a laboratory method for disassembling clusters of eggs that float or remain grouped as a raft, allowing researchers to handle each egg independently. In practice, controlled mechanical manipulation under liquid loosens contacts between neighboring eggs while minimizing damage to the embryo and preserving viability. This makes individual eggs available for synchronized culture, controlled crosses, developmental observation, and genotype- or phenotype-based selection in genetic studies. By improving the separation and tracking of embryos from a shared raft, the method supports reproducible experiments involving inheritance, embryonic development, and laboratory population management.

An in vitro method to mimic in vivo epithelial differentiation is described. Many viruses target epithelial cells as part of their viral life cycle, and this method provides a means of examining virus:host interactions that more closely resembles that which occurs in vivo. This technique can be used with primary keratinocytes, established cell lines, as well as normal or diseased biopsy tissue.

An efficient, three-step synthesis of RAFT-based fluorescent glycopolymers, consisting of glycomonomer preparation, copolymerization, and post-modification, is demonstrated. This protocol can be used to prepare RAFT-based statistical glycopolymers with desired structures.

A technique to probe the lipid raft partitioning of fluorescent proteins at the plasma membrane of living cells is described. It takes advantage of the disparity in diffusion times of proteins located inside or outside of lipid rafts. Acquisition can be performed dynamically in control conditions or after drug addition.
