Blood Cell Isolation

Blood cell isolation is a laboratory technique used to separate specific blood cell populations from whole blood for study and analysis. It commonly relies on differences in cell density, size, surface markers, or adherence, using methods such as density-gradient centrifugation, magnetic separation, or fluorescence-activated cell sorting. In immunology and infection research, isolated leukocytes, including lymphocytes and monocytes, support investigations of immune activation, pathogen interactions, cytokine responses, and cellular function. Consistent isolation improves sample quality and enables downstream assays such as flow cytometry, microscopy, cell culture, and molecular analysis, helping researchers characterize host defenses and disease-related changes.

Here we describe a protocol for isolating subsets of precursor B-cells from umbilical cord blood. A sufficient quantity and quality of nucleic acids may be extracted from the cells and used in subsequent assays utilizing DNA or RNA.
