Brewer's Thioglycollate

Brewer's Thioglycollate is an enriched reducing culture medium used to grow and maintain microorganisms, particularly anaerobic bacteria that are inhibited by oxygen. Sodium thioglycollate and other reducing components lower the oxidation-reduction potential, while the medium's small amount of agar limits oxygen diffusion and helps establish an oxygen gradient; resazurin can indicate oxygen exposure through a color change. In immunology and infection research, this medium supports the recovery, enrichment, and study of anaerobic pathogens from clinical or environmental specimens. It also helps investigators assess oxygen requirements, maintain bacterial cultures, and examine microbial growth under conditions that more closely resemble oxygen-limited sites in the body.

Here, we present a modified TGA method for estimation of lignin content in herbaceous plant biomass. This method estimates the lignin content by forming specific thioether bonds with lignin and presents an advantage over the Klason method, as it requires a relatively small sample for lignin content estimation.

Bathophenanthroline Sulfonate-Based Colorimetric Assay: A Simple and Rapid Method for Quantitation of Non-Heme Iron in Mouse Liver Tissue

2025

In this video, we describe a colorimetric-based method to determine non-heme iron content in mouse liver tissue. Bathophenanthroline sulfonate is used as the chromogenic substrate in the presence of a reducing agent, which converts ferric iron to chelatable ferrous iron.

We describe simultaneous mechanical testing and 3D-imaging of the arterial wall of isolated, live human resistance arteries, and Fiji and Ilastik image analyses for the quantification of elastin and collagen spatial organization and volume densities. We discuss the use of these data in mathematical models of arterial wall mechanics.
