Germ-free Chickens

Germ-free chickens are birds hatched and maintained under controlled, sterile conditions without detectable microorganisms, providing a model for separating host biology from microbial influences. Researchers use sterilized eggs, isolators, and monitored feed and housing to prevent colonization, then introduce defined microbes or pathogens under controlled conditions. This system helps reveal how the microbiota shapes immune development, intestinal function, metabolism, and resistance to infection. In immunology and infection research, germ-free chickens support precise comparisons between uncolonized, conventionally raised, and selectively colonized birds, improving understanding of host-microbe interactions and informing studies of vaccines, probiotics, disease mechanisms, and poultry health.

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Auditory brainstem neurons of avians and mammals are specialized for fast neural encoding, a fundamental process for normal hearing functions. These neurons arise from distinct precursors of embryonic hindbrain. We present techniques utilizing electroporation to express genes in the hindbrain of chicken embryos to study gene function during auditory development.
