Imaging Flow Cytometer

An imaging flow cytometer is an analytical instrument that combines the high-throughput measurement of flow cytometry with the visual detail of fluorescence microscopy, enabling simultaneous analysis of thousands of individual cells. As cells flow through a focused stream, the instrument captures brightfield and fluorescence images in multiple channels, while software quantifies markers, morphology, localization, and cell-to-cell variation. In immunology and infection research, this approach supports immune-cell phenotyping, identification of infected cells, intracellular pathogen detection, and analysis of host-pathogen interactions. By linking molecular signals with cellular structure, imaging flow cytometry provides statistically robust insights that conventional microscopy or nonimaging cytometry alone may miss.

Spaceflight blood diagnostics need innovation. Few demonstrations have been published illustrating in-flight, reduced-gravity health diagnostic technology. Here we present a method for construction and operation of a parabolic flight test rig for a prototype point-of-care flow-cytometry design, with components and preparation strategies adaptable to other setups.

The micronucleus (MN) assay is a well-established test for quantifying DNA damage. However, scoring the assay using conventional techniques such as manual microscopy or feature-based image analysis is laborious and challenging. This paper describes the methodology to develop an artificial intelligence model to score the MN assay using imaging flow cytometry data.

We review our recent results on the integration of fluorescent microscopy and imaging flow cytometry tools on a cell-phone using compact and cost-effective opto-fluidic attachments. These cell-phone based micro-analysis devices might be useful for cytometric analysis, such as performing various cell counting tasks as well as for high-throughput screening of e.g., water samples in resource limited settings.
