Macrophage Detection

Macrophage detection comprises methods used to identify and characterize macrophages in tissues, cell cultures, and biological samples, making it essential for studying innate immunity and infection. These methods combine cellular morphology with macrophage-associated surface or intracellular markers, often measured by immunostaining, flow cytometry, or analysis of phagocytic activity, to distinguish macrophages from other immune cells and assess their activation state. In this video, we demonstrate a cytotoxicity assay to assess Cryptococcus neoformans, a pathogenic fungus, infection in macrophages. The cytotoxicity is determined by the release of cytoplasmic lactate dehydrogenase enzyme due to macrophage lysis caused by Cryptococcus neoformans infection.

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.

The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice.
