Oxygen Availability

Oxygen availability describes the amount of oxygen accessible to cells and tissues, a condition that regulates metabolism and immune function. In infected tissues, inflammation, vascular changes, and microbial consumption create oxygen gradients; low oxygen stabilizes hypoxia-inducible factor transcription factors, reshaping glycolysis, cytokine production, leukocyte behavior, and pathogen metabolism. Measuring or manipulating oxygen availability in culture systems and infection models helps researchers reproduce tissue microenvironments, understand host-pathogen interactions, and assess how hypoxia influences antimicrobial defense, pathogen persistence, and treatment response. These studies can clarify why immune responses differ across anatomical sites and support models and therapies that account for oxygen-dependent biology.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
