Viral Rna Transcription

Viral RNA transcription is the process by which viruses produce messenger RNA and other RNA molecules needed to express their genes and replicate within infected cells. Depending on the viral genome, a virus may use a host RNA polymerase or encode an RNA-dependent RNA polymerase that copies RNA templates into complementary strands, often generating subgenomic mRNAs for individual proteins. These reactions occur in specialized replication complexes and can be regulated by genome structure, polymerase activity, and host-cell conditions. Understanding viral RNA transcription clarifies how infections progress, how infected cells trigger innate immune responses, and how antiviral drugs can target viral polymerases or RNA synthesis.

Using Click Chemistry to Measure the Effect of Viral Infection on Host-Cell RNA Synthesis

This method describes the use of click chemistry to measure changes in host cell transcription after infection with the Rift Valley fever virus (RVFV) strain MP-12. Results can be visualized qualitatively via fluorescence microscopy or obtained quantitatively through flow cytometry. This method is adaptable for use with other viruses.

Real-Time Quantitative Reverse Transcription PCR for Diagnosing Viral Infections

This video demonstrates a quantitative method for detecting viral infection using real-time reverse transcription PCR. The reverse transcription step converts the RNA to cDNA, which is then amplified via PCR cycles. The presence of viral RNA in the sample is confirmed by analyzing the amplification and dissociation curves of DNA obtained from the PCR.

Detection of Viral RNA by Fluorescence in situ Hybridization (FISH)

A fluorescence in situ hybridization (FISH) method was developed to visually detect viral genomic RNA using fluorescence microscopy. A probe is made with specificity to the viral RNA that can then be identified using a combination of hybridization and immunofluorescence techniques. This technique offers the advantage of identifying the localization of the viral RNA or DNA at steady-state, providing information on the control of intracellular virus trafficking events.
