Automated Cell Culture

Automated cell culture is the use of robotic instruments, software, and controlled laboratory equipment to grow and maintain cells with limited manual intervention. These systems coordinate tasks such as media dispensing, cell seeding, incubation, feeding, passaging, and imaging under defined temperature, gas, and timing conditions, while standardized workflows improve consistency between cultures. In medicine, automated cell culture supports drug screening, disease modeling, toxicity testing, and the production of cells for research and therapeutic development. By increasing throughput, reducing handling variability, and enabling continuous monitoring, automation helps researchers generate more reproducible data and scale complex cell-based studies.

The goal of this protocol is to manufacture pathogen-specific clinical-grade T cells using a bench-top, automated, second generation cell enrichment device that incorporates a closed cytokine capture system and does not require dedicated staff or use of a GMP facility. The cytomegalovirus pp65-specific-T cells generated can be directly administered to patients.

Peptide adsorption to plasticware during traditional tip-based serial dilutions can significantly impact potency determination and confound the understanding of structure-activity relationships used for lead identification and lead optimization phases of drug discovery. Here methods for automated acoustic non-contact serial dilution of peptide samples are described.
