Cell Purity Assessment

Cell purity assessment is the evaluation of how closely a cell preparation matches its intended cellular composition, an essential step in medical research, diagnostics, and cell-based therapies. It typically combines morphological examination with marker-based methods, such as immunostaining or flow cytometry, to identify target cells and detect unwanted cell types, while viability and contamination tests assess sample quality. Reliable purity measurements support reproducible experiments, accurate disease models, and safer therapeutic products. Standardized assessment also helps monitor cell isolation, culture, and manufacturing processes, improving quality control and enabling consistent interpretation of clinical and research outcomes.

Here we describe a robust method of determining immune cell identity and purity through epigenetic signatures detected using quantitative PCR (qPCR). DNA demethylation at a specific locus serves as a unique identifier for a particular cell type and allows for identification of CD8+, regulatory, or Th17 T cells.
