Cocktail Aon Approach

The Cocktail AON Approach is a therapeutic strategy that combines multiple antisense oligonucleotides (AONs) to address genetic variation within a disease rather than targeting a single mutation. Each AON binds a complementary sequence in pre-messenger RNA and can alter splicing, redirecting exon inclusion or skipping to help produce a functional protein. In medicine, this approach is particularly relevant to genetically heterogeneous disorders such as Duchenne muscular dystrophy, where different AONs may target distinct exons or mutation classes. By broadening molecular coverage, AON cocktails can support more adaptable, mutation-specific treatment development and may inform future precision therapies.

Multi-exon Skipping Using Cocktail Antisense Oligonucleotides in the Canine X-linked Muscular Dystrophy

Exon skipping is currently a most promising therapeutic option for Duchenne muscular dystrophy (DMD). To expand the applicability for DMD patients and to optimize the stability/function of the resulting truncated dystrophin proteins, a multi-exon skipping approach using cocktail antisense oligonucleotides was developed and we demonstrated systemic dystrophin rescue in a dog model.

Whole blood Immunophenotyping is indispensable for monitoring the human immune system. However, the number of reagents required and the instability of specific fluorochromes in premixed cocktails necessitates daily reagent preparation. Here we show that semi-automated preparation of staining antibody cocktail helps establish reliable immunophenotyping by minimizing variability in reagent dispensing.

GENPLAT (GLBRC Enzyme Platform) is an automated platform for discovery and optimization of enzyme cocktails for biomass degradation. It can be adapted to multiple feedstocks and mixtures of enzymes containing multiple components.

Promotion of Survival and Differentiation of Neural Stem Cells with Fibrin and Growth Factor Cocktails after Severe Spinal Cord Injury

Fibrin matrices containing growth factors were used to retain grafted neural stem cells into sites of complete spinal cord transection. Grafted cells completely filled the lesion cavity and differentiated into multiple neural cell types, including neurons that extended axons into host spinal cord over long distances.
