Lad Occlusion

LAD occlusion is a blockage of the left anterior descending coronary artery, an important vessel that supplies blood to the front of the heart and much of the interventricular septum. It most often develops when an atherosclerotic plaque ruptures and triggers thrombus formation, sharply reducing oxygen delivery and potentially causing an anterior myocardial infarction. Clinicians assess suspected occlusion using symptoms, electrocardiography, cardiac biomarkers, and coronary angiography. Rapid restoration of blood flow, commonly through percutaneous coronary intervention, can limit myocardial injury, preserve cardiac function, and improve survival, making early recognition and treatment a central concern in emergency medicine.

This article presents a unique closed-chest technique for inducing myocardial ischemia-reperfusion injury (IRI) in mice. The presented method allows mice to breathe spontaneously while remotely inducing myocardial ischemia. This provides access to the animal for studying the dynamic processes of ischemia and reperfusion in situ and in real-time via noninvasive imaging.

We provide a reliable method for left anterior descending artery (LAD) ligation in a mouse model. This method is comparatively less invasive than other methods, involving endotracheal intubation, a left-sided thoracotomy approach, and thoracentesis. This method can be used as a model for both acute and chronic myocardial infarction (MI).

We describe a highly reproducible method for the permanent occlusion of a rodent major cerebral blood vessel. This technique can be accomplished with very little peripheral damage, minimal blood loss, a high rate of long-term survival, and consistent infarct volume commensurate with the human clinical population.
