Multiplexed Serological Testing

Multiplexed serological testing is a laboratory method that measures antibodies, antigens, or other immune markers for multiple targets in a single patient sample, providing broader information than a single-analyte assay. The method uses distinct capture reagents, often arranged on coded beads or a spatial array, to bind target molecules simultaneously; labeled detection reagents and instrument-based signal measurement then identify and quantify each reaction. In medicine, multiplexed serological testing can support differential diagnosis, infection surveillance, vaccine and immune-response studies, and evaluation of autoimmune or inflammatory patterns. By conserving sample volume and comparing related markers in parallel, it can improve testing efficiency while requiring careful validation for cross-reactivity and interpretation.

Multiplex fluorescence immunoassays are becoming a reference for serology testing. Their application, however, is limited by the complexity and time-consuming bead-analyte coupling processes. This report presents a semi-automated, reproducible multiplex microsphere bead coupling protocol that is comparable to the standard method while considerably reducing time to results.

A three-channel dual-reporter fluorescence flow analysis system was used to develop a bead-based multiplex immunoassay that simultaneously evaluates serum samples for IgG and IgM elicited against multiple antigens of different Borrelia species that cause Lyme borreliosis in Europe and North America.

Using Luminex Corporation’s xMAP microsphere technology, we have developed the Multiplexed Fluorometric ImmunoAssay (MFIA) for serosurveillance of various laboratory animal species. The MFIA is a suspension microarray where antigen, tissue control or immunoglobulins are covalently linked to color-coded polystyrene microspheres. The MFIA testing method as well as various troubleshooting topics is addressed.

The serological pipette is frequently used in the laboratory for transferring milliliter volumes of liquid, from less than 1 ml to up to 50 ml. The pipettes can be sterile, plastic, and disposable or sterilizable, glass and reusable. Both kinds of pipettes use a pipet-aid, for the aspiration and dispensation of liquids. When working in a laboratory, it is imperative to minimize sources of contamination. Aseptic technique refers to procedures that permit transfer of cultures and reagents while avoiding contact with non-sterile surfaces. Serological pipettes and micropipettors are used to measure precise volumes without compromising sterility of solutions used in experiments.
