Streptozotocin Model

The Streptozotocin model is an experimental disease model used to induce diabetes in laboratory animals, enabling investigation of hyperglycemia, insulin deficiency, and diabetic complications. Streptozotocin selectively enters pancreatic beta cells through glucose transporter 2, where it causes DNA alkylation and oxidative stress, leading to beta-cell injury and reduced insulin production; dose and administration conditions influence disease severity. Researchers use this model to study diabetes pathophysiology, evaluate glucose-lowering therapies, and assess complications affecting organs such as the kidneys, nerves, and blood vessels. Its reproducibility makes it a valuable tool in preclinical medicine, although findings require careful interpretation because the induced disease may not fully replicate human diabetes.

A Zebrafish Model of Diabetes Mellitus and Metabolic Memory

Metabolic memory is the phenomenon by which diabetic complications persist and progress unimpeded even after euglycemia is achieved pharmaceutically. Here we describe a diabetes mellitus zebrafish model which is unique in that it allows for the examination of the mitotically transmissible epigenetic components of metabolic memory in vivo.

Mouse Model for Pancreas Transplantation Using a Modified Cuff Technique

Among abdominal solid organ transplantation, pancreatic grafts are prone to develop severe ischemia reperfusion injury-associated graft damage, leading eventually to early graft loss. This protocol describes a model of murine pancreas transplantation using a non-suture cuff technique, ideally suited for analyzing these early, deleterious damages.

In Vivo Modeling of the Morbid Human Genome using Danio rerio

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.

Here we describe a novel diabetic murine model utilizing hairless mice for real-time, non-invasive, monitoring of biofilm wound infections of bioluminescent Pseudomonas aeruginosa. This method can be adapted to evaluate infection of other bacterial species and genetically modified microorganisms, including multi-species biofilms, and test the efficacy of antibiofilm strategies.
