Subcutaneous Adipose Arterioles

Subcutaneous adipose arterioles are small resistance blood vessels that regulate arterial blood flow into fat tissue beneath the skin, making them important for tissue perfusion and metabolic function. Their vascular smooth muscle changes vessel diameter in response to neural, hormonal, and local metabolic signals, thereby controlling blood pressure, oxygen delivery, and exchange of nutrients and waste products within adipose tissue. Studying these arterioles helps clarify how obesity, diabetes, inflammation, and impaired vascular function alter adipose biology. Their responses also provide clinically relevant insight into thermoregulation, wound healing, and the vascular mechanisms underlying metabolic disease.

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.

We standardized an abdominal adipose tissue biopsy using a non-diathermy method performed under local anesthesia. Three cases of excessive post-operation bleeding out of 115 operations (2.61%) occurred. We conclude that an abdominal adipose tissue surgical biopsy using a non-diathermy method can be safely applied to healthy men.

This manuscript describes a simple and reproducible protocol for isolation of intracerebral arterioles (a group of blood vessels encompassing parenchymal arterioles, penetrating arterioles and pre-capillary arterioles) from mice, to be used in pressure myography, immunofluorescence, biochemistry, and molecular studies.

2024

Here, we describe a protocol for isolating, culturing, and phenotyping microvascular endothelial cells from human subcutaneous adipose tissue (hSATMVECs). Additionally, we describe an experimental model of hSATMVEC-adipocyte cross-talk.
