Three-dimensional Matrix

A three-dimensional matrix is a spatially organized network of biological or synthetic material that provides structural support and biochemical cues for cells, tissues, or engineered constructs. Its interconnected architecture controls cell adhesion, migration, proliferation, and differentiation by presenting binding sites, regulating mechanical forces, and permitting the transport of nutrients and signaling molecules. In medicine, three-dimensional matrices are used in tissue engineering, wound repair, organoid culture, drug testing, and regenerative research, where they can model tissue microenvironments more realistically than flat cultures. Matrix composition, stiffness, porosity, and degradation rate help determine biological outcomes and guide the design of patient-relevant therapies.

Engineering Three-dimensional Epithelial Tissues Embedded within Extracellular Matrix

This manuscript describes a soft lithography-based technique to engineer uniform arrays of three-dimensional (3D) epithelial tissues of defined geometry surrounded by extracellular matrix. This method is amenable to a wide variety of cell types and experimental contexts and allows for high-throughput screening of identical replicates.

The bone extracellular matrix (BEM) model for osteosarcoma (OS) is well established and shown here. It can be used as a suitable scaffold for mimicking primary tumor growth in vitro and providing an ideal model for studying the histologic and cytogenic heterogeneity of OS.

Explants from the midbrain dopamine system and striatum are used in a collagen matrix assay for the in vitro analysis of mesostriatal and striatonigral pathway development. In this assay axonal outgrowth and guidance can be manipulated and quantified. It can also be modified for assessing other regions or molecular cues.

Cellular processes such as cell migration have traditionally been studied on two-dimensional, stiff plastic surfaces. This report describes a technique for directly visualizing protein localization and analyzing protein dynamics in cells migrating in a more physiologically relevant, three-dimensional matrix.
