Anti-fade Medium

Anti-fade medium is a chemical mounting medium that preserves fluorescence in labeled specimens during microscopy, helping researchers obtain stable, interpretable images. It works by limiting photobleaching, in which excitation light drives reactive chemical reactions that damage fluorescent molecules; antioxidant or free-radical-scavenging components in the medium reduce this loss of signal and often improve sample preservation. In neuroscience, anti-fade media support imaging of fluorescent proteins, immunolabeled neurons, synaptic markers, and other structures in fixed brain tissue. By extending fluorescence stability, they enable more reliable visualization, comparison, and quantitative analysis of cellular organization and neural connectivity.

This protocol describes a novel and efficient method to quickly initiate operant responding for ethanol in rats that, contrary to standard methods, does not require water deprivation or saccharin/sucrose fading to initiate responding.

Using a Macrophage Conditioned Medium to Promote Neuron Extensions

This video demonstrates an assay for promoting the growth of neuron projections known as neurite outgrowth using the macrophage-conditioned medium. Co-culture neurons and macrophages with cAMP and harvest the macrophage-derived conditioned medium. Growing the neurons with the conditioned medium promotes neurite outgrowths. These outgrowths are fluorescently labeled and visualized using a fluorescence microscope.
