Dual-mouse Tracking

Dual-mouse tracking is a behavioral neuroscience method that simultaneously records and analyzes the movements of two mice, enabling quantitative study of social interactions and coordinated behavior. Typically, overhead video is processed with computer-vision or pose-estimation algorithms that identify each animal, follow body landmarks across frames, and calculate measures such as position, speed, orientation, distance, and contact. These data reveal patterns of approach, avoidance, investigation, and behavioral synchrony under controlled conditions. By linking social behavior with neural recordings, stimulation, genetic manipulations, or disease models, dual-mouse tracking helps researchers investigate the circuits and mechanisms underlying communication, affiliation, aggression, and other interactions.

This protocol provides a reliable method to establish humanized mice with both human immune system and liver cells. Dual reconstituted immunodeficient mice achieved via intrasplenic injection of human hepatocytes and CD34+ hematopoietic stem cells are susceptible to human immunodeficiency virus-1 infection and recapitulate liver damage as observed in HIV-infected patients.

Monocytes are key regulators of innate immunity and play a critical role in the renewal of the peripheral mononuclear phagocytic system and in case of inflammation. This manuscript describes the procedure of real time imaging of the mouse calvaria bone marrow to study the monocyte mobilisation mechanism.

Locomotor activity (LMA) is a simple and easily performed measurement of behavior in mice. Coupling of video tracking software (VTS) and LMA allows for the improvement of specificity and sensitivity, especially when compared with the manual, line crossing method of LMA analysis. Additionally VTS allows long-term tracking of mouse LMA.
