Nerve Stump Dissection

Nerve stump dissection is a microsurgical technique used to expose, isolate, and examine the severed end of a peripheral nerve, providing insight into injury responses and regeneration. During the procedure, surrounding connective tissue is carefully removed while preserving the nerve, allowing researchers to distinguish the proximal stump, which retains connections to the cell body, from the distal stump and its degenerating fibers. In neuroscience, this approach supports the study of Wallerian degeneration, axonal sprouting, neuroma formation, and nerve repair. Findings can help evaluate injury mechanisms, regenerative responses, and experimental treatments designed to restore peripheral nerve function.

Here, we describe a method for loading a calcium-sensitive dye through the frog nerve stump into the nerve endings. We also present a protocol for the recording and analysis of fast calcium transients in the peripheral nerve endings.

This article describes a technique to insert a hollow conduit between the spinal cord stumps after complete transection and fill with Schwann cells (SCs) and injectable basement membrane matrix in order to bridge and promote axon regeneration across the gap.

Dissection of Pelvic Autonomic Ganglia and Associated Nerves in Male and Female Rats

The major pelvic ganglia contain parasympathetic and sympathetic neurons that innervate pelvic organs. Here we describe a dissection method and provide schematics for identification of these ganglia and their associated nerves. These methods can be applied to experimental manipulation of these ganglia in vivo or removal post-mortem for further study.

Although the peripheral nervous system (PNS) is capable of significant repair after injury, little is known about the cellular and molecular mechanisms that govern this phenomenon. Using live, transgenic zebrafish and a reproducible nerve transection assay, we can study dynamic glial cell behaviors during nerve degeneration and regeneration.
