Periodic Acid Oxidation

Periodic acid oxidation is a chemical method that detects carbohydrates and other compounds containing adjacent hydroxyl groups, making it valuable for visualizing carbohydrate-rich structures in biological tissues. Periodic acid cleaves carbon-carbon bonds between neighboring hydroxyl groups, generating aldehydes that can react with Schiff reagent in the periodic acid–Schiff (PAS) procedure to produce a colored signal. In neuroscience, this approach helps identify glycogen, glycoproteins, glycolipids, and basement-membrane components in nervous tissue, supporting the study of cellular organization, tissue pathology, and neurodegenerative changes. Its compatibility with histological sections also makes it useful for correlating molecular composition with microscopic anatomy.

De novo lipogenesis and β-fatty acid oxidation constitute key metabolic pathways in hepatocyte, pathways that are perturbed in several metabolic disorders, including fatty liver disease. Here we demonstrate isolation of mouse primary hepatocytes and describe quantification of β-fatty acid oxidation and lipogenesis.
