Th Egfp Transgene

The TH-EGFP transgene is a genetic reporter construct that links expression of enhanced green fluorescent protein (EGFP) to tyrosine hydroxylase (TH), an enzyme associated with catecholamine synthesis. Regulatory sequences from the TH gene drive EGFP production in TH-expressing cells, causing dopaminergic and noradrenergic neurons to fluoresce and become identifiable in nervous-system tissue. In neuroscience, this transgene supports visualization, targeted recording, cell isolation, and circuit mapping of catecholaminergic populations. By providing a fluorescent marker for genetically defined neurons, it helps researchers examine neuronal development, connectivity, activity, and disease-related changes in relevant brain circuits.

This video demonstrates the generation of neural progenitor cells from non-transgenic and transgenic human pluripotent stem cells (hPSCs). hPSCs are differentiated into neural progenitor cells using a neuronal differentiation medium. While differentiation is induced in non-transgenic cells using small molecules, the transgenic hPSCs are differentiated by adding an antibiotic that induces the promoter for a neuronal differentiation-specific transcription factor.

Transgenic worms are commonly used in C. elegans research. Described is a simple, yet effective, protocol to introduce transgenes into worms using biolistic bombardment with DNA-coated gold particles. The effort involved and results of bombardment compare favorably with microinjection for the generation of transgenic animals.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.
