Live Cell Confocal

Live-cell confocal microscopy is a fluorescence imaging technique that captures optical sections of living cells, allowing researchers to observe molecular and cellular behavior over time. A focused laser excites fluorescent labels within the specimen, while a pinhole blocks out-of-focus light so the detector records sharper images from a defined focal plane; sequential sections can be combined into three-dimensional views. In biochemistry, this method helps track protein localization, organelle dynamics, membrane trafficking, and intracellular signaling in their native cellular context. Time-lapse imaging can reveal dynamic responses to chemical treatments or changing conditions, although experiments must balance image quality with photobleaching and phototoxicity.

Cell Death Associated with Abnormal Mitosis Observed by Confocal Imaging in Live Cancer Cells

The cytotoxic activity of the phenanthridine PJ-34 in cancer cells undergoing mitosis was documented in real time by live confocal imaging. PJ-34 eradicated human breast cancer MDA-MB-231 cells harboring extra-centrosomes in mitosis. Unlike normal bi-focal mitosis, the extra-centrosomes were not clustered in the two spindle poles in the presence of PJ-34.

Several methods are available for measuring intracellular pH but very few of these allow simultaneous measurement of cytoplasmic and organellar pH. Here, we describe in detail a rapid and accurate methodology to simultaneously measure cytoplasmic and vesicular pH by ratiometric imaging of living cells.
