Muscle Graft Preparation

Muscle graft preparation is the controlled processing of muscle tissue for transplantation, reconstruction, or use in engineered tissue constructs. The process involves collecting and handling the graft under sterile conditions, removing unsuitable material, and shaping or conditioning the tissue while preserving its structural organization and cellular viability. In bioengineering, careful preparation supports integration with the host environment and can promote restoration of muscle form and function. These grafts are used to study tissue repair, develop regenerative therapies, and evaluate biomaterials or culture conditions designed to improve muscle regeneration and long-term performance.

Preparation of Light-responsive Membranes by a Combined Surface Grafting and Postmodification Process

A plasma-induced polymerization procedure is described for the surface-initiated polymerization on polymer membranes. Further postmodification of the grafted polymer with photochromic substances is presented with a protocol of conducting permeability measurements of light-responsive membranes.

The Mouse Cremaster Muscle Preparation for Intravital Imaging of the Microcirculation

A tissue preparation is described for visualization and experimental manipulation of the living microcirculation. In anesthetized male mice, the thin, highly vascularized cremaster muscle is prepared for intravital microscopy to study microvascular networks including arterioles, capillaries and venules. This preparation is readily adapted for rats and hamsters.

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.
