Liposome Reconstitution

Liposome reconstitution is a biological technique that incorporates membrane proteins, peptides, or other biomolecules into artificial lipid vesicles to recreate selected features of a cell membrane. It typically involves forming liposomes from phospholipids, removing detergents or other solubilizing agents, and allowing the membrane components to assemble into a stable bilayer, sometimes with controlled orientation or encapsulation. This approach provides a defined system for studying membrane transport, receptor signaling, enzyme activity, and protein–lipid interactions. By isolating specific components from the complexity of living cells, liposome reconstitution supports mechanistic research, assay development, drug delivery studies, and the design of biomimetic membranes.

Biological membrane fusion is catalyzed by specialized fusion proteins. Measuring the fusogenic properties of proteins can be achieved by lipid mixing assays. We present a method for purifying recombinant Drosophila atlastin, a protein that mediates homotypic fusion of the ER, reconstituting it to preformed liposomes, and testing for fusion capacity.

Described here is a rapid and effective procedure for functional reconstitution of purified wild-type and mutant CFTR protein that preserves activity for this chloride channel, which is defective in Cystic Fibrosis. Iodide efflux from reconstituted proteoliposomes mediated by CFTR allows studies of channel activity and the effects of small molecules.

Depletion and Reconstitution of Macrophages in Mice

Macrophages play a central role in homeostasis and pathology in many tissues. The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice. These techniques allow determination of the role that polarized macrophages play in health and disease.
