Bmp Signal Activation

Bmp signal activation is the process by which bone morphogenetic protein (BMP) ligands initiate intracellular signaling to regulate cell behavior, making it central to development, tissue formation, and homeostasis. BMPs bind paired type I and type II serine/threonine kinase receptors, allowing receptor-mediated phosphorylation of SMAD1/5/8; these proteins associate with SMAD4 and move into the nucleus to control target-gene expression, while additional noncanonical pathways can contribute to the response. Studying this pathway helps explain cell-fate decisions, pattern formation, and organ development, and supports research into skeletal disorders, tissue repair, cancer, and strategies for controlling stem-cell differentiation.

2023

Optogenetic manipulation of signaling pathways can be a powerful strategy to investigate how signaling is decoded in development, regeneration, homeostasis, and disease. This protocol provides practical guidelines for using light-oxygen-voltage sensing domain-based Nodal and bone morphogenic protein (BMP) signaling activators in the early zebrafish embryo.

MALT1 regulates innate immunity but how this occurs remains ill-defined. We used the selective MALT1 paracaspase inhibitor MLT-827 to unravel the contribution of MALT1 to innate signaling downstream of Toll-like or C-type lectin-like receptors, demonstrating that MALT1 regulates the production of myeloid cytokines, and downstream of C-type lectin-like receptors, selectively.

Photostimulation by Femtosecond Laser Activates Extracellular-signal-regulated Kinase (ERK) Signaling or Mitochondrial Events in Target Cells

2019

Tightly-focused femtosecond laser can deliver precise stimulation to cells by being coupled into a confocal microscopy enabling the real-time observation and photostimulation. The photostimulation can activate cell molecular events including ERK signaling pathway and mitochondrial flashes of reactive oxygen species.

Mimicking the Function of Signaling Proteins: Toward Artificial Signal Transduction Therapy

2016

We present guidelines for developing synthetic 'chemical transducers' that can induce communication between naturally unrelated proteins. In addition, detailed protocols are presented for synthesizing and testing a specific 'transducer' that enables a growth factor to activate a detoxifying enzyme and consequently, to regulate the cleavage of an anticancer prodrug.
