Nuclei Isolation

Nuclei isolation is the laboratory process of separating cell nuclei from the surrounding cytoplasm and other cellular components for molecular and structural analysis. It typically uses controlled mechanical homogenization to disrupt cell membranes while preserving the nuclear envelope, followed by filtration and differential centrifugation or density-gradient centrifugation to enrich intact nuclei. The isolated nuclei can support DNA, chromatin, and nuclear RNA analyses, including sequencing and imaging, across diverse biological samples. In biology research, this approach enables cell-type profiling, epigenetic studies, gene-regulation analysis, and investigation of nuclear organization while reducing interference from cytoplasmic material.
