Genomic Rnai Library

A genomic RNAi library is a collection of sequence-specific RNA interference reagents designed to reduce the expression of many or all genes in an organism or cell system, enabling systematic gene-function analysis. Each double-stranded RNA, small interfering RNA, or short hairpin RNA contains a sequence complementary to a target transcript; after processing, the guide strand directs the RNA-induced silencing complex to the matching messenger RNA, promoting its degradation or suppressing translation. Researchers use these libraries in high-throughput loss-of-function screens to identify genes involved in development, signaling, disease pathways, and cellular responses, linking genetic perturbations to measurable biological phenotypes.

Large Insert Environmental Genomic Library Production

Construction of a fosmid library with environmental genomic DNA isolated from the vertical depth continuum of a seasonally hypoxic fjord is described. The resulting clone library is picked into 384-well plates and archived for downstream sequencing and functional screening by the application of an automated colony picking system.

RNAi in C. elegans

RNA interference (RNAi) is a widely used technique in which double stranded RNA is exogenously introduced into an organism, causing knockdown of a target gene. In the nematode, C. elegans, RNAi is particularly easy and effective because it can be delivered simply by feeding the worms bacteria that express double stranded RNA (dsRNA) that is complementary to a gene of interest. First, this video will introduce the concept of RNA interference and explain how it causes targeted gene knockdown.

Quantitative and Automated High-throughput Genome-wide RNAi Screens in C. elegans

We describe a protocol using C. elegans and RNAi feeding libraries that allows automated measurement of multiple parameters such as fluorescence, size and opacity of individual worms in a population. We give one example of a screen to identify genes involved in anti-fungal innate immunity in C. elegans.
