Osteoblast Formation

Osteoblast formation is the biological process by which progenitor cells differentiate into osteoblasts, the bone-forming cells responsible for producing new bone matrix. In bone biology, mesenchymal stem or stromal cells commit to the osteogenic lineage through signals such as Wnt, then activate transcription factors including RUNX2 and osterix that establish osteoblast identity. Mature osteoblasts secrete collagen-rich osteoid and promote its mineralization with calcium phosphate, while some become osteocytes or bone-lining cells. Studying this process clarifies skeletal development and bone remodeling and supports research into fracture healing, osteoporosis, biomaterials, and regenerative therapies.

In this video, we describe a method for the detection of osteoblast-secreted calcium deposits using an anionic dye, Alizarin Red S.
