Sub-pixel Resolution

Sub-pixel resolution is an imaging capability that estimates the position or structure of biological features more precisely than the physical size of a camera or detector pixel. It works by analyzing how light from an object spreads across neighboring pixels, then using signal intensity, optical models, interpolation, or repeated measurements to estimate its location within a pixel. In biological imaging, this approach can improve measurements of molecular movement, cell boundaries, organelle dynamics, and fluorescent labels without requiring smaller detector pixels. Sub-pixel methods support quantitative microscopy and image registration, although their accuracy depends on factors such as signal-to-noise ratio, optical calibration, sampling, and specimen stability.

Test Samples for Optimizing STORM Super-Resolution Microscopy

We describe the preparation of three test samples and how they can be used to optimize and assess the performance of STORM microscopes. Using these examples we show how to acquire raw data and then process it to acquire super-resolution images in cells of approximately 30-50 nm resolution.

High Resolution Phonon-assisted Quasi-resonance Fluorescence Spectroscopy

The manuscript describes a method of phonon-assisted quasi-resonant fluorescence spectroscopy that incorporates both laser-limited resolution and photoluminescence (PL) spectroscopy. This method utilizes optical phonons to provide linewidth-limited resolution spectra of atom-like semiconductor structures in the energy domain. The method is also easily realized with a single spectrometer optical spectroscopy setup.
