Fibroblast-derived Matrix

Fibroblast-derived matrix is an extracellular matrix (ECM) produced and deposited by fibroblasts, providing a biologically relevant scaffold for studying cell behavior. In culture, fibroblasts secrete and organize proteins such as collagen and fibronectin; researchers can then remove the cells, leaving an acellular matrix that preserves tissue-like structural and biochemical cues. In cancer research, this matrix models elements of the tumor microenvironment and helps investigators examine how ECM composition and organization influence cancer-cell adhesion, spreading, migration, invasion, and responses to treatment. It also supports comparisons between normal and cancer-associated fibroblast matrices, clarifying how stromal remodeling contributes to tumor progression.

Fibroblast-Derived 3D Matrix System Applicable to Endothelial Tube Formation Assay

The aim of this method is to obtain fibroblast-derived 3D matrices as a natural scaffold for subsequent cellular assays. Fibroblasts are seeded in a pre-treated culture plate and stimulated with ascorbic acid for matrix generation. Matrices are decellularized and blocked to culture relevant cells (e.g., endothelial cells).

The extracellular matrix plays a major role in defining the microenvironment of cells and in modulating cell behavior and phenotype. We describe a rapid method for the isolation of cell-derived extracellular matrix, which can be adapted to different scales for microscopic, biochemical, proteomic, or functional studies.

Robust derivation of human induced pluripotent stem (hiPS) cells was achieved by using non-integrating Sendai virus (SeV) vector mediated reprogramming of dermal fibroblasts. hiPS cell maintenance and clonal expansion was performed using xeno-free and chemically defined culture conditions with recombinant human laminin 521 (LN-521) matrix and Essential E8 (E8) Medium.

Generation of induced pluripotent stem cells provides fascinating prospects for the derivation of autologous transplants. However, progression through a pluripotent state and laborious re-differentiation still hinders clinical translation. Here we describe the derivation of adult human fibroblasts and their direct conversion into induced neural progenitor cells and the subsequent differentiation into neural lineages.
