Metastatic Factor Identification

Metastatic factor identification is the systematic discovery and characterization of genes, proteins, cell states, and microenvironmental signals that enable cancer cells to spread from a primary tumor to distant organs. Researchers combine genomic and transcriptomic profiling with functional assays to determine whether candidate factors regulate processes such as epithelial–mesenchymal transition, invasion, intravasation, survival in circulation, extravasation, or colonization of new tissue. In cancer research, these studies can reveal biomarkers associated with metastatic risk and expose molecular dependencies that support tumor dissemination. Validating such factors in cell models, animal systems, and patient samples helps prioritize targets for prognostic testing and therapies designed to prevent or treat metastatic disease.

We present a protocol to demonstrate a novel somatic gene transfer system utilizing RIP-Tag; RIP-tva mouse model to study the function of genes in metastasis. The avian retroviruses are delivered intracardiacally to ensure gene transfer into pre-malignant, noninvasive lesions of pancreatic β cells in adult mice.
