Tfh Cells

Tfh cells, or follicular helper T cells, are specialized CD4+ lymphocytes that support B-cell maturation and antibody production, making them important regulators of adaptive immunity and potential contributors to tumor immunity. They migrate into B-cell follicles through the chemokine receptor CXCR5, maintain a follicular program regulated by Bcl-6, and stimulate B cells through contact-dependent signals such as ICOS and cytokines including IL-21. In cancer research, Tfh cells are studied within tumors and tertiary lymphoid structures, where they may shape antitumor antibody responses, influence immune-cell organization, and serve as indicators of treatment response or targets for immunotherapeutic strategies.

The bm12 Inducible Model of Systemic Lupus Erythematosus (SLE) in C57BL/6 Mice

The transfer of bm12 lymphocytes into a C57BL/6 recipient is an established model of systemic lupus erythematosus. Here we describe how to initiate disease using this model and how to characterize T follicular helper cells, germinal center B cells and plasma cells by flow cytometry.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.
