Supplemental Oxygen

Supplemental oxygen is the clinical administration of oxygen at concentrations or flow rates above those found in ambient air to prevent or treat inadequate tissue oxygenation. Delivered through devices such as nasal cannulas, face masks, or mechanical ventilators, it raises the oxygen concentration available for inhalation, increasing alveolar oxygen and supporting diffusion into the blood. Clinicians use supplemental oxygen for conditions including hypoxemia caused by respiratory disease, acute illness, or impaired gas exchange, while monitoring oxygen saturation and clinical response. Appropriate titration can improve organ function and reduce physiological stress, whereas unnecessary or excessive oxygen may cause harm in some patients.

The goal of this protocol is to develop a model system for the effect of hyperoxia on cystic fibrosis airway microbial communities. Artificial sputum medium emulates the composition of sputum, and hyperoxic culture conditions model the effects of supplemental oxygen on lung microbial communities.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
