Embryonic Cardiotoxicity

Embryonic cardiotoxicity is the harmful effect of chemical, pharmaceutical, or environmental exposures on the developing heart during embryogenesis, a critical concern in developmental biology and prenatal safety. These agents can disrupt cardiac progenitor specification, heart tube formation, electrical signaling, contractility, or vascular development through processes such as oxidative stress, altered ion-channel activity, and interference with developmental signaling pathways. Researchers assess these effects using animal models, embryonic stem cell-derived cardiomyocytes, and organoid or embryo-like systems to measure changes in morphology, rhythm, beating, and cell survival. Such studies help identify teratogenic hazards, clarify mechanisms of congenital heart defects, and improve drug screening and risk assessment.

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.

Embryonic Stem Cells

2019

Embryonic stem (ES) cells are undifferentiated pluripotent cells, meaning they can produce any cell type in the body. This gives them tremendous potential in science and medicine since they can generate specific cell types for use in research or to replace body cells lost due to damage or disease. Origin ES cells are present in the inner cell mass of an embryo at the blastocyst stage, which occurs at about 3–5 days after fertilization in humans before the embryo is implanted in the uterus.

Immunostaining of Dissected Zebrafish Embryonic Heart

A rapid way to conduct immunostaining of zebrafish embryonic heart is described. Compared to the whole mount immunostaining approach, this method dramatically increases the penetration of the antibodies, which allows obtaining high resolution images that reveal cellular/subcellular structures in the heart within a much reduced processing time.
