Hemogenic Program Induction

Hemogenic program induction is the activation of developmental pathways that give selected endothelial cells the capacity to produce blood-forming cells, a key transition in embryonic hematopoiesis. It works by coordinating signaling cues and transcriptional regulators that establish hemogenic endothelium and drive the endothelial-to-hematopoietic transition, during which cells change identity, detach from the vessel wall, and acquire hematopoietic properties. In developmental biology, this process helps explain how the blood system emerges from vascular tissues. Reproducing it in pluripotent stem cells supports studies of blood development, disease modeling, and efforts to generate clinically useful hematopoietic cells in vitro.

The protocol described here details the induction of a hemogenic program in mouse embryonic fibroblasts via overexpression of a minimal set of transcription factors. This technology may be translated to the human system to provide platforms for future study of hematopoiesis, hematologic disease, and hematopoietic stem cell transplant.

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.

Chen, PhD, Department of Physics & Astronomy, College of Science, Purdue University, West Lafayette, IN This experiment will use inductive coils to demonstrate the concept of inductor and inductance. Magnetic induction will be demonstrated using a rod magnet inserted into or extracted away from the core of a coil to induce a transient electromotive force (emf) voltage in the coil, measured by a voltmeter. This experiment will also demonstrate the mutual inductance between...

Programmed electrical stimulation provides the ability to determine conduction properties of the heart, and the possibility to induce and terminate cardiac arrhythmias using various pacing protocols. Using a transvenous catheter, intracardiac electrogram recordings can be obtained in mice following programmed electrical stimulation protocols to identify arrhythmogenic substrates.
