Hypocotyl Pinching

Hypocotyl pinching is a plant biology technique that applies controlled mechanical pressure to the hypocotyl, the stem-like region of a seedling between the root and cotyledons, to create a reproducible injury. By compressing or constricting tissues, it disrupts transport and activates wound-response pathways, including local signaling that supports tissue repair and vascular reconnection. Researchers use this assay to examine how seedlings respond to physical damage, environmental stress, and altered growth conditions. Results can reveal mechanisms of regeneration, resilience, and resource allocation, helping connect plant development with environmental adaptation.

Pinching-off of Coated Vesicles

2023

Vesicle budding is orchestrated by distinct cytosolic proteins such as adaptor proteins, coat proteins, and GTPases. To initiate vesicle budding, membrane-bending proteins containing crescent-shaped BAR domains bind to the lipid heads in the bilayer and distort the membrane to form a protein-coated vesicle bud. Adaptors proteins such as AP2 for clathrin-coated vesicles can nucleate on the deformed membrane. Finally, coat proteins such as clathrin or COPI and COPII assemble into a coat forming...

This article describes the methods for screening the genes controlling plasmodesmal permeability and hence auxin gradient during tropic response. This includes the measurement of the degree of tropic response in hypocotyl of Arabidopsis thaliana and checking plasmodesmal permeability by 8-hydroxypyrene-1,3,6-trisulfonic acid (HPTS) loading and finally callose level assessment.
