Cytotoxic T Cell Functionality

Cytotoxic T cell functionality is the capacity of CD8+ T lymphocytes to identify and eliminate infected, abnormal, or cancerous cells, making it central to adaptive immunity and infection control. After the T cell receptor recognizes a specific peptide presented by major histocompatibility complex class I, coordinated signaling triggers formation of an immunological synapse and release of perforin and granzymes, or engagement of Fas-mediated apoptosis. Assessing these responses through target-cell killing, cytokine production, and degranulation assays helps characterize immune protection, disease progression, vaccine responses, and immunotherapies.

Radial Mobility and Cytotoxic Function of Retroviral Replicating Vector Transduced, Non-adherent Alloresponsive T Lymphocytes

We describe a protocol to monitor radial mobility of non-adherent immune cells in vitro using a cell sedimentation manifold/slide apparatus. Cell migration is tracked on monolayers of tumor cells or on extracellular matrix proteins. Examination by light and fluorescence microscopy allows for observation of cell mobility and cytotoxic functionality.

Quantitative High-throughput Single-cell Cytotoxicity Assay For T Cells

We describe a single-cell high-throughput assay to measure cytotoxicity of T cells when incubated with tumor target cells. This method employs a dense, elastomeric array of sub-nanoliter wells (~100,000 wells/array) to spatially confine the T cells and target cells at defined ratios and is coupled to fluorescence microscopy to monitor effector-target conjugation and subsequent apoptosis.
